"ds_process_way": "1. Experimental Design\r\n\r\nThis study was conducted on the northern slope of the central Kunlun Mountains (36°10′-36°50′N, 80°10′-80°50′E), where seven sampling transects were established along an elevation gradient ranging from 1,707 m to 3,548 m. These included desert (1,707 m), montane semi-shrub desert (1,960 m, 2,448 m, and 2,746 m), montane desert steppe (2,905 m), and alpine steppe (3,248 m and 3,548 m) ecosystems (Table 1). During the sampling period, no vegetation was observed at the 1,707 m site. At elevations between 1,960 m and 2,746 m, vegetation was predominantly composed of shrubs and semi-shrubs, including Calligonum roborovskii, Reaumuria soongonica, and Sympegma regelii. The vegetation at 2,905 m belonged to montane desert steppe and was mainly composed of Stipa capillata, Seriphidium rhodanthum, and Achnatherum splendens. At 3,548 m, the vegetation was classified as alpine steppe, with Stipa purpurea, Kobresia filifolia, and Potentilla species as the dominant plants.\r\n\r\n2. Sampling Methods\r\n\r\nAccording to vegetation type, four independent quadrats (1 m × 1 m) were established at elevations of 3,548 m, 3,248 m, and 2,905 m, with a horizontal distance of 10 m between adjacent quadrats. At elevations of 2,746 m, 2,448 m, 1,960 m, and 1,707 m, four independent large quadrats (10 m × 10 m) were established, with a horizontal spacing of 100 m between quadrats. Within each quadrat, plant diversity and vegetation cover were surveyed, and aboveground vegetation biomass was collected.\r\n\r\nAfter removing surface litter, five topsoil cores (0-10 cm) were randomly collected along the diagonal line of each quadrat. These soil cores were thoroughly mixed to form one composite sample. In total, 28 soil samples were collected from quadrats distributed across the seven elevation levels.\r\n\r\nDuring soil sampling, disposable gloves were worn at all times, and the soil auger was sterilized with 75% ethanol before collecting each sample. After transportation to the laboratory, soil samples from different elevations were sieved through a 2 mm mesh that had been sterilized under ultraviolet light for 30 min to prevent cross-contamination. Subsequently, each soil sample was divided into two subsamples: one portion was air-dried for physicochemical analyses, while the other was stored at −20°C for subsequent extraction of soil microbial DNA.\r\n\r\n3. Analytical Procedures\r\n\r\nSoil DNA was extracted using the E.Z.N.A.® Soil DNA Kit. DNA concentration was measured with a TBS-380 fluorometer, DNA purity was assessed using a NanoDrop 2000 spectrophotometer, and DNA integrity was evaluated by 1% agarose gel electrophoresis.\r\n\r\nThe extracted DNA was fragmented to approximately 400 bp using a Covaris M220 ultrasonicator. Paired-end sequencing libraries were then constructed and subjected to metagenomic sequencing on the Illumina NovaSeq PE150 platform. The sequencing data have been deposited in the Genome Sequence Archive (GSA) of the National Genomics Data Center under accession number PRJCA017392.\r\n\r\nRaw sequencing reads were quality-filtered using Fastp and assembled with MEGAHIT. Open reading frames (ORFs) were predicted using Prodigal, and genes with lengths ≥100 bp were retained and translated into amino acid sequences. Carbon cycle-related functional genes were annotated against the KEGG database. A non-redundant gene catalog was constructed using SOAPaligner clustering at 95% sequence identity.\r\n\r\nCAZyme functional annotation was performed using HMMER, and the identified carbohydrate-active enzymes were classified into six enzyme families: glycoside hydrolases (GHs), glycosyltransferases (GTs), carbohydrate esterases (CEs), carbohydrate-binding modules (CBMs), polysaccharide lyases (PLs), and auxiliary activities (AAs). Gene abundance was normalized using the RPKM method. The α-diversity and β-diversity of functional genes were calculated, and dominant microbial taxa were taxonomically annotated using Diamond searches against the NR database.\r\n\r\nAll statistical analyses were conducted in R version 4.2.1. One-way analysis of variance (ANOVA) followed by Duncan's multiple range test was used to compare differences in functional genes, microbial communities, and environmental factors among elevations. The Shannon index was used to evaluate α-diversity, while linear regression analysis was applied to examine relationships among elevation, diversity, and functional gene abundance. Principal Coordinates Analysis (PCoA) and Permutational Multivariate Analysis of Variance (PERMANOVA) were used to assess differences in community composition and enzyme composition.\r\n\r\nFurthermore, redundancy analysis (RDA), variance partitioning analysis (VPA), Pearson correlation analysis, and Mantel tests were employed to reveal the relationships among environmental factors, microbial communities, and carbon cycle functional genes. Nonlinear regression analysis was also conducted to evaluate the response of the MAOC/POC ratio to environmental factors.",
